Sex-specific consequences of host shift for morphology and fluctuating asymmetry in a seed beetle: an experimental evolution approach

AB  - Morphological plasticity may be induced by altering host plants and commonly it is sexspecific in phytophagous insects. It is hypothesized that stress in insects caused by a host shift
leads to morphological changes and developmental destabilization, which may be identified
by fluctuating asymmetry. We performed reciprocal transplant experiments in eight replicated
populations of the seed beetle Acanthoscelides obtectus (Say) (Coleoptera: Chrysomelidae,
Bruchinae) adapted to common bean (Phaseolus vulgaris L., Fabaceae) as its ancestral host
and chickpea (Cicer arietinum L., Fabaceae) as a suboptimal host. Using methods of
geometric morphometrics we assessed the effects of the short- and long-term host shift on size
and shape variation of beetles of both sexes. We also tested the hypothesis that fluctuating
asymmetry is lower when the beetles develop within seeds of the ancestral host. Our results
showed that females respond more plastically related to their body size and shape of the
abdomen, particularly during a short-term host shift, suggesting that females have a greater
importance in maintaining the population on a new host. The level of fluctuating asymmetry
is lowest in males that have evolved for generations on bean indicating that they have the
most canalized development, so we suggested that symmetry in A. obtectus is maintained by
sexual selection. Even if the beetles are adapted to a suboptimal host over many generations,
they stabilize their development after returning to the ancestral host. Our work indicates that
host shifts may change morphological aspects of the beetles in a sex-specific manner and
consequently influence their developmental trajectories.

Impact of untreated wastewaters on the microbiological water quality of the Danube River and its tributaries in Serbia

AB  - Pollution of surface waters still represents one of the major environmental challenges in the Republic of Serbia, where wastewaters are discharged directly into the recipients without proper treatment. Within the last decade, we have assessed the microbiological water quality of more than 100 sites situated at different water bodies in Serbia. Quality was determined using faecal indicator bacteria, while microbial source tracking was employed to assess the source of pollution. Almost 50 % of the investigated sites were characterized by critical or even higher level of faecal contamination indicating that untreated wastewaters indeed represent significant pollution pressure on surface waters. Human-associated markers were prevalent in samples from the majority of contaminated sites but the source of pollution was not exclusively human-associated. 
The impact on the water quality of the Danube River was demonstrated in our previous research conducted at the whole river level within the Joint Danube Surveys. In the river stretch from Novi Sad to its confluence with the Velika Morava River, all the midstream samples were critically polluted. In this section, the highest level of pollution was recorded downstream of Belgrade. As ultimate recipients of wastewaters, Danube and its largest tributary Sava currently represent the only solution for disposing of wastewaters originating from the Serbian capital‘s 1,700,000 inhabitants. Such kind of disposal rises additional issues such as antimicrobial resistance and presence of infectious agents in water. For instance during COVID-19 pandemic, we have demonstrated that SARS-CoV-2 RNA can be detected even in surface waters of the Danube River at the sites receiving high wastewater loads from Belgrade which was the unique case for the Basin. 
Despites its extreme importance in this case, wastewater-based epidemiology is neglected in our country and hereby we would like to emphasize the need for implementation of program of such kind in Serbia.

AB  - Without being aware of their chemical composition, many cultures have used herbaceous peony roots for medicinal purposes. Modern phytopreparations intended for use in human therapy require specific knowledge about the chemistry of peony roots and their biological activities. In this study, ethanol–water extracts were prepared by maceration and microwave- and ultrasound-assisted extractions (MAE and UAE, respectively) in order to obtain bioactive molecules from the roots of Paeonia tenuifolia L., Paeonia peregrina Mill., and Paeonia officinalis L. wild growing in Serbia. Chemical characterization; polyphenol and flavonoid content; antioxidant, multianti-enzymatic, and antibacterial activities of extracts; and in vitro gastrointestinal digestion (GID) of hot water extracts were performed. The strongest anti-cholinesterase activity was observed in PT extracts. The highest anti-ABTS (2,2′-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) radical potential was observed in PP extracts, whereas against DPPH (2,2-diphenyl-1-picrylhydrazyl radicals), the best results were achieved with PO extracts. Regarding antibacterial activity, extracts were strongly potent against Bacillus cereus. A molecular docking simulation was conducted to gather insights into the binding affinity and interactions of polyphenols and other Paeonia-specific molecules in the active sites of tested enzymes. In vitro GID of Paeonia teas showed a different recovery and behavior of the individual bioactives, with an increased recovery of methyl gallate and digallate and a decreased recovery of paeoniflorin and its derivatives. PT (Gulenovci) and PP (Pirot) extracts obtained by UAE and M were more efficient in the majority of the bioactivity assays. This study represents an initial step toward the possible application of Paeonia root extracts in pharmacy, medicine, and food technologies.

What is a B chromosome? Early definitions revisited

We reconsidered early definitions in light of recently published studies. While there are many characteristics that vary among different B chromosomes, such as their evolutionary origins, size, segregation behaviors, gene content, and function, there is one defining trait of all B chromosomes: they are nonessential for the organism. The points raised here may be useful for framing future B chromosome studies and help guide the categorization of new chromosomal elements that are uncovered in genomic studies.

The prevalence of Borrelia burgdorferi sensu lato in Apodemus species, the most common small mammal in Belgrade forests

AB  - Lyme disease is a widespread zoonosis and a major health concern in Europe. This zoonosis is caused by tick-transmitted pathogenic Gram-negative spirochetes belonging to a group called Borrelia burgdorferi sensu lato (s.l.). The Borrelia transmission cycle includes the vector, vertebrate reservoir, and host species. At least five different genospecies have been identified as human pathogens in Europe. Previous studies showed that the most frequently detected genospecies in Belgrade is Borrelia afzelii, whose main reservoirs are small mammals. The most common small mammals in Belgrade forests are mice from the genus Apodemus (A. flavicollis and A. sylvaticus), which are infective for ticks throughout their lifespan with varying efficiency.
We investigated the prevalence of B. burgdorferi s.l. in Apodemus species in six fragment urban forests in the Belgrade area, and Avala Mt. A total of 142 animals were analysed for the presence of B. burgdorferi s.l. in isolated DNA by RT-PCR. For positive samples, we performed PCR and sequencing of the OspA gene to identify the genospecies. 
Our study revealed varying prevalence rates of only one genospecies, B. afzelii, in Apodemus specimens across different localities. The highest prevalence was observed on Avala mountain (25%), followed by Miljakovačka šuma (16%), Kvantaš (9%), Ada Ciganlija (9%) and Zvezdara (8%). No infected animals were detected in Stepin Gaj and Košutnjak.
This is the study of Borrelia prevalence in small mammal reservoirs in the Belgrade area. Understanding the prevalence of B. burgdorferi s.l. in the genus Apodemus in the Belgrade area is of great value for comprehending the ecology of this pathogen and potential risk zones.
AB  - Лајмска болест је широко распрострањена зооноза и данас представља велики здравствени проблем у Европи. Ову зоонозу изазивају патогене грам-негативне спирохете које припадају групи која се зове Borrelia burgdorferi sensu lato (s.l.) и преносе се крпељима. Цео циклус преноса борелије укључује векторе, кичмењаке као резервоар и врсте крајњег домаћина. Најмање пет различитих геноврста идентификоване су као људски патогени у Европи. Досадашња истраживања су показала да је најчешће забележена геноврста у Београду Borrelia afzelii, чији су главни резервоари мали сисари. Најчешћи мали сисари у београдским шумама су мишеви из рода Apodemus (A. flavicollis and A. sylvaticus) који су, са различитом ефикасношћу, током целог свог животног века инфективни за крпеље.
Истраживали смо распрострањеност B. burgdorferi s.l. у врстама Apodemus у шест великих урбаних шума на подручју Београда и на планини Авали. Укупно 142 животиње су анализиране на присуство B. burgdorferi s.l. у изолованој ДНК помоћу РТ-ПЦР. За позитивне узорке, урадили  смо ПЦР и секвенцирање OspA гена да бисмо идентификовали геноврсте.
Наша студија је открила различите стопе преваленције само једне геноврсте, B. afzelii, у јединкама рода Apodemus, на различитим локалитетима. Највећа преваленција забележена је на планини Авали (25%), затим Миљаковачкој шуми (16%), Кванташу (9%), Ади Циганлији (9%) и Звездари (8%). У Степином Гају и Кошутњаку није откривена ни једна заражена животиња.
Ово је студија о распрострањености борелије у малим сисарима као резервоарима на подручју Београда. Разумевање распрострањености B. burgdorferi s.l. у роду Apodemus на подручју Београда је од великог значаја за сагледавање екологије овог патогена и потенцијалних ризичних зона.

This zoonosis is caused by tick-transmitted pathogenic Gram-negative spirochetes belonging to a group called Borrelia burgdorferi sensu lato (s.l.). The Borrelia transmission cycle includes the vector, vertebrate reservoir, and host species. At least five different genospecies have been identified as human pathogens in Europe. Previous studies showed that the most frequently detected genospecies in Belgrade is Borrelia afzelii, whose main reservoirs are small mammals. The most common small mammals in Belgrade forests are mice from the genus Apodemus (A. flavicollis and A. sylvaticus), which are infective for ticks throughout their lifespan with varying efficiency.
We investigated the prevalence of B. burgdorferi s.l. in Apodemus species in six fragment urban forests in the Belgrade area, and Avala Mt. A total of 142 animals were analysed for the presence of B. burgdorferi s.l. in isolated DNA by RT-PCR. For positive samples, we performed PCR and sequencing of the OspA gene to identify the genospecies. 
Our study revealed varying prevalence rates of only one genospecies, B. afzelii, in Apodemus specimens across different localities. The highest prevalence was observed on Avala mountain (25%), followed by Miljakovačka šuma (16%), Kvantaš (9%), Ada Ciganlija (9%) and Zvezdara (8%). No infected animals were detected in Stepin Gaj and Košutnjak.
This is the study of Borrelia prevalence in small mammal reservoirs in the Belgrade area. AB  - We have recently characterized experimental autoimmune encephalomyelitis (EAE) induced in DA rats with spinal cord homogenate without complete Freund’s adjuvant (CFA). The main advantage of this multiple sclerosis model is the lack of CFA-related confounding effects which represent the major obstacles in translating findings from EAE to multiple sclerosis. Here, antigen specificity of the cellular and humoral immune response directed against the central nervous system was explored. The reactivity of T and B cells to myelin basic protein, myelin oligodendrocyte glycoprotein, and β-synuclein was detected. Having in mind that reactivity against β-synuclein was previously associated with autoimmunity against the brain, the infiltration of immune cells into different brain compartments, i.e. pons, cerebellum, hippocampus, and cortex was determined. T cell infiltration was observed in all structures examined. This finding stimulated investigation of the effects of immunization on DA rat behavior using the elevated plus maze and the open field test. Rats recovered from EAE displayed increased anxiety-like behavior. These data support CFA-free EAE in DA rats as a useful model for multiple sclerosis research.

AB  - Diets high in fat and sugar lead to metabolic syndrome (MetS) and related chronic diseases. We
investigated the effects of commercially available, cold-pressed polyphenol-rich black currant
(BC) and cornelian cherry (CC) juices on the prevention of MetS in Wistar rats induced by a 10-
weeks high-fat high-fructose (HFF) diet. Juice consumption, either BC or CC, with a HFF diet
resulted in lower serum triglycerides compared to only the HFF consumption. Both juices also
mitigated the effects of HFF on the liver, pancreas, and adipose tissue, by preserving liver and
pancreas histomorphology and reducing visceral fat and adipocyte size. Furthermore, supple-
mentation with both juices reduced glucagon and up-regulated insulin expression in the pancreas
of the rats on the HFF diet, whereas the BC also showed improved glucose regulation. BC juice
also reduced the expression of IL-6 and hepatic inflammation compared to the group only on HFF
diet. Both juices, especially BC, could be a convenient solution for the prevention of MetS in
humans.

AB  - Parkinson’s disease (PD) is a chronic, age-related, progressive multisystem disease associated with neuroinflammation and immune dysfunction. This review discusses the methodological
approaches used to study the changes in central and peripheral immunity in PD, the advantages and
limitations of the techniques, and their applicability to humans. Although a single animal model
cannot replicate all pathological features of the human disease, neuroinflammation is present in
most animal models of PD and plays a critical role in understanding the involvement of the immune
system (IS) in the pathogenesis of PD. The IS and its interactions with different cell types in the
central nervous system (CNS) play an important role in the pathogenesis of PD. Even though culture
models do not fully reflect the complexity of disease progression, they are limited in their ability to
mimic long-term effects and need validation through in vivo studies. They are an indispensable tool
for understanding the interplay between the IS and the pathogenesis of this disease. Understanding
the immune-mediated mechanisms may lead to potential therapeutic targets for the treatment of PD.
We believe that the development of methodological guidelines for experiments with animal models
and PD patients is crucial to ensure the validity and consistency of the results.

AB  - Our study was aimed to investigate the effects of commercially available, cold-pressed polyphenol-rich black currant (BC) juice on fat accumulation, blood lipids, insulin resistance, high blood pressure, and inflammation, in rats placed on high-fat high-fructose (HFF) diet. Coadministration of BC juice significantly reduced diet-induced adiposity and plasma triglycerides levels, improved glucose tolerance, and suppressed hepatic inflammation during prolonged HFF feeding in rats. These findings suggest that consumption of BC juice may  counter the harmful effects of the Western diet rich in fats and sugars, and to be useful for the prevention of metabolic syndrome development and progression.

AB  - The aim of the study was to test the significance of the EPT index in the water quality assessment of three types of water bodies in hilly and mountainous region of Serbia. The aquatic macroinver-tebrate community was dominated by the group of insects, of which 95 taxa represent the EPT group. We compared the obtained values of biological indices used for the assessment of water quality according to the national legislation with the overall status assessment represented by the ecological quality classes (EQC). The results of the Spearman correlation test showed a negative correlation of EQC with the EPT index, BMWP score, H', total number of taxa and number of sen-sitive taxa, while a positive correlation was observed for the values of SI and Tubificinae %. The values of EQC and biological indices were subjected to principal component analysis (PCA). The results showed that the parameters that contributed most to the differences were the EPT index, the BMWP score and the number of sensitive taxa. The results indicate that the EPT index is an excellent indicator of changes in water quality and an important tool for the ecological categoriza-tion of water bodies in mountain regions.

AB  - Studying the membrane physiology of filamentous fungi is key to understanding their interactions with the environment and crucial for developing new therapeutic strategies for disease-causing pathogens. However, their plasma membrane has been inaccessible for a micron-sized patch-clamp pipette for pA current recordings due to the rigid chitinous cell wall. Here, we report the first femtosecond IR laser nanosurgery of the cell wall of the filamentous fungi, which enabled patch-clamp measurements on protoplasts released from hyphae. A reproducible and highly precise (diffraction-limited, submicron resolution) method for obtaining viable released protoplasts was developed. Protoplast release from the nanosurgery-generated incisions in the cell wall was achieved from different regions of the hyphae. The plasma membrane of the obtained protoplasts formed tight and high-resistance (GΩ) contacts with the recording pipette. The entire nanosurgical procedure followed by the patch-clamp technique could be completed in less than 1 hour. Compared to previous studies using heterologously expressed channels, this technique provides the opportunity to identify new ionic currents and to study the properties of the ion channels in the protoplasts of filamentous fungi in their native environment.

Background Carrot is an important vegetable crop grown worldwide. The major economic problem in carrot cultivation is yellow disease caused by Bactericera trigonica, which induces biotic stress and has the greatest impact on crop productivity. Comprehensive studies on the mechanism of carrot defense response to biotic stress caused by B. trigonica infestation have yet to be conducted.
Methods The changes in photosynthetic pigments, proline, TPC, H2O2 and MDA content, DPPH radical scavenging ability, and antioxidant enzyme activity of SOD, CAT, and POX in carrot leaves in response to insect sex (female and male), rapid response (during the frst six hours), and long-term response to B. trigonica infestation were evaluated.
Results The results of our study strongly suggest that B. trigonica infestation causes signifcant changes in primary and secondary metabolism and oxidative status of carrot leaves. Photosynthetic pigment content, TPC, and DPPH and CAT activities were signifcantly reduced in carrot leaves in response to insect infestation. On the other hand, proline, H2O2 content, and the activity of the antioxidant enzymes superoxide dismutase and peroxidase were increased in carrot leaves after B. trigonica infestation. The results indicate that B. trigonica attenuates and delays the oxidative stress responses of carrot, allowing long-term feeding without visible changes in the plant. Carrot responded to longterm B. trigonica infestation with an increase in SOD and POX activity, suggesting that these enzymes may play a key
role in plant defense mechanisms.
Conclusions This is the frst comprehensive study strongly suggesting that B. trigonica infestation causes signifcant changes in primary and secondary metabolism and an attenuated ROS defense response in carrot leaves that enables long-term insect feeding. The information provides new insights into the mechanisms of carrot protection against B. trigonica infestation.

AB  - Two novel hybrid compounds, CON1 and CON2, have been developed by combining sclareol (SC) and doxorubicin (DOX) into a single molecular entity. These hybrid compounds have a 1:1 molar ratio of covalently linked SC and DOX. They have demonstrated promising anticancer properties, especially in glioblastoma cells, and have also shown potential in treating multidrug-resistant (MDR) cancer cells that express the P-glycoprotein (P-gp) membrane transporter. CON1 and CON2 form nanoparticles, as confirmed by Zetasizer, transmission electron microscopy (TEM), and chemical modeling. TEM also showed that CON1 and CON2 can be found in glioblastoma cells, specifically in the cytoplasm, different organelles, nucleus, and nucleolus. To examine the anticancer properties, the U87 glioblastoma cell line, and its corresponding multidrug-resistant U87-TxR cell line, as well as patient-derived astrocytoma grade 3 cells (ASC), were used, while normal human lung fibroblasts were used to determine the selectivity. CON1 and CON2 exhibited better resistance and selectivity profiles than DOX, showing less cytotoxicity, as evidenced by real-time cell analysis, DNA damage determination, cell death induction, mitochondrial respiration, and mitochondrial membrane depolarization studies. Cell cycle analysis and the β-galactosidase activity assay suggested that glioblastoma cells die by senescence following CON1 treatment. Overall, CON1 and CON2 showed great potential as they have better anticancer features than DOX. They are promising candidates for additional preclinical and clinical studies on glioblastoma.

Classification of sustainability potential of genetic resources of local grapevine varieties in Serbia

AB  - The starting point for every viticultural and wine-producing country
with respect to local grapevine varieties is their identification, inventory, preservation and
development of genetic resources of those varieties. There are currently 224 grapevine varieties cultivated in Serbia for the purpose of commercial production of grapes and wine.
Out of that number, 31 wine varieties are local. Vineyards under those varieties can be differentiated by their importance for production of grapes and wine, and by the level of their
endangerment, that is, sustainability in conditions caused by climate changes. This paper
presents the creation, that is, the modeling of the Method for Vineyard Sustainability Classification (MVSC). The purpose of MVSC classification is: to valorize vineyards with local
wine grapevine varieties on grounds of their endangerment and sustainability, based on 20
examined and categorized individual vineyard sustainability parameters; to carry out comprehensive classification into one of the four established vineyard sustainability classes
(Class A – very endangered vineyards, Class B – endangered vineyards, Class C – sustainable
vineyards and Class D – very sustainable vineyards); spatial identification and presentation
of vineyards based on determined vineyard sustainability class through application of GIS
technology; and finally, application of Network Analysis (NA), prioritization of examined
parameters and, therefore, vineyards. A total of 10,402 vineyards under local grapevine wine
varieties were used for modeling, and it was determined that 29 vineyards with the total
surface of 1.2 hectares should be classified in Class A, while 2,883 vineyards with the total
surface of 158.2 hectares should be classified in Class B. With respect to the strength of 20
individual vineyard sustainability parameters, it was determined that the parameter Structure
of the vine rootstock (SVR) has the greatest impact, and priority in selection of vineyards in
different sustainability classes should be given to vineyards without rootstocks. In accordance
with the scientific justification of obtained results, the MVSC enables comprehensive classification of the potential for sustainability of genetic resources of local grapevine varieties
in Serbia, and it can be applied in other countries and wine-growing areas, as well as to
other groups of grapevine varieties

AB  - Acute respiratory distress syndrome (ARDS) became a focus of intensive research due to its death toll during the Covid-19 pandemic. An uncontrolled and excessive inflammatory response mediated by proinflammatory molecules such as high mobility group box protein 1 (HMGB1), IL-6, and TNF mounts as a response to infection. In this study, ethyl pyruvate (EP), a known inhibitor of HMGB1, was tested in the model of murine ARDS induced in C57BL/6 mice by intranasal administration of polyinosinic:polycytidylic (poly(I:C)). Intraperitoneal administration of EP ameliorated the ARDS-related histopathological changes in the lungs of poly(I:C)-induced ARDS and decreased numbers of immune cells in the lungs, broncho-alveolar lavage fluid and draining lymph nodes (DLN). Specifically, fewer CD8+ T cells and less activated CD4+ T cells were observed in DLN. Consequently, the lungs of EP-treated animals had fewer damage-inflicting CD8+ cells and macrophages. Additionally, the expression and production of proinflammatory cytokines, IL-17, IFN-γ and IL-6 were downregulated in the lungs. The expression of chemokine CCL5 which recruits immune cells into the lungs was also reduced. Finally, EP downregulated the expression of HMGB1 in the lungs. Our results imply that EP should be further evaluated as a potential candidate for ARDS therapy.

AB  - We have recently characterized experimental autoimmune encephalomyelitis (EAE) induced in DA rats with spinal cord homogenate without complete Freund’s adjuvant (CFA). The main advantage of this multiple sclerosis model is the lack of CFA-related confounding effects which represent the major obstacles in translating findings from EAE to multiple sclerosis. Here, antigen specificity of the cellular and humoral immune response directed against the central nervous system was explored. The reactivity of T and B cells to myelin basic protein, myelin oligodendrocyte glycoprotein, and β-synuclein was detected. Having in mind that reactivity against β-synuclein was previously associated with autoimmunity against the brain, the infiltration of immune cells into different brain compartments, i.e. pons, cerebellum, hippocampus, and cortex was determined. T cell infiltration was observed in all structures examined. This finding stimulated investigation of the effects of immunization on DA rat behavior using the elevated plus maze and the open field test. Rats recovered from EAE displayed increased anxiety-like behavior. These data support CFA-free EAE in DA rats as a useful model for multiple sclerosis research.

AB  - Simple and scalable synthetic approach was used for the preparation of thirteen novel tacrine derivatives consisting of tacrine and N-aryl-piperidine-4-carboxamide moiety connected by a five-methylene group linker. An anti-Alzheimer disease (AD) potential of newly designed tacrine derivatives was evaluated against two important AD targets, acetylcholinesterase (AChE) and butyrylcholinesterase (BuChE). In vitro pharmacological evaluation showed strong ChE inhibitory activity of all compounds, with IC50 values ranging from 117.5 to 455 nM for AChE and 34 to 324 nM for BuChE. As a representative of the series with the best cytotoxicity / ChE inhibitory activity ratio, expressed as the selectivity index (SI), 2-chlorobenzoyl derivative demonstrated mixed-type inhibition on AChE and BuChE, suggesting binding to both CAS and PAS of the enzymes. It also exhibited antioxidant capacity and neuroprotective potential against amyloid-β (Aβ) toxicity in the culture of neuron-like cells. In-depth computational analysis corroborated well with in vitro ChE inhibition, illuminating that all compounds exhibit significant potential in targeting both enzymes. Molecular dynamics (MD) simulations revealed that 2-chlorobenzoyl derivative, created complexes with AChE and BuChE that demonstrated sufficient stability throughout the observed MD simulation. Computationally predicted ADME properties indicated that these compounds should have good blood–brain barrier (BBB) permeability, an important factor for CNS-targeting drugs. Overall, all tested compounds showed promising pharmacological behavior, highlighting the multi-target potential of 2- chlorobenzoyl derivative which should be further investigated as a new lead in the drug development process.

AB  - The invasion of non-native plant species has a detrimental effect on native biodiversity. In a seven-year research project, we investigated the occurrence of the invasive species Asclepias syriaca L.in the Deliblato Sands protected area, located at a south-eastern part of the Pannonian Plain, and identified the factors that contribute significantly to its colonisation. The distribution of this invasive species was monitored on more than 300 km of the accessory, bordering and selected internal roads. A. syriaca occurs within the protected area but is much more widespread on accessory and bordering roads. The number of locations of A. syriaca increased every year of the study, even within the protected area, indicating a further spread of this species. A. syriaca is much more abundant on the north-eastern edge than in the south-west. The reason for this is most likely the much larger area of abandoned agricultural land in the north-east. Roads used for public transport are the main entry points for the further spread of A. syriaca. In contrast, recreational trail use does not enhance the spread in the Deliblato Sands natural reserve. This study can be used to further analyse the ecological dynamics of A. syriaca and to develop timely strategies by which to prevent or slow down its spread.

An anti-Alzheimer disease (AD) potential of newly designed tacrine derivatives was evaluated against two important AD targets, acetylcholinesterase (AChE) and butyrylcholinesterase (BuChE). In vitro pharmacological evaluation showed strong ChE inhibitory activity of all compounds, with IC50 values ranging from 117.5 to 455 nM for AChE and 34 to 324 nM for BuChE. As a representative of the series with the best cytotoxicity / ChE inhibitory activity ratio, expressed as the selectivity index (SI), 2-chlorobenzoyl derivative demonstrated mixed-type inhibition on AChE and BuChE, suggesting binding to both CAS and PAS of the enzymes. It also exhibited antioxidant capacity and neuroprotective potential against amyloid-β (Aβ) toxicity in the culture of neuron-like cells. In-depth computational analysis corroborated well with in vitro ChE inhibition, illuminating that all compounds exhibit significant potential in targeting both enzymes. Molecular dynamics (MD) simulations revealed that 2-chlorobenzoyl derivative, created complexes with AChE and BuChE that demonstrated sufficient stability throughout the observed MD simulation. Computationally predicted ADME properties indicated that these compounds should have good blood–brain barrier (BBB) permeability, an important factor for CNS-targeting drugs. Overall, all tested compounds showed promising pharmacological behavior, highlighting the multi-target potential of 2- chlorobenzoyl derivative which should be further investigated as a new lead in the drug development process.

A Geometric Morphometrics Approach to the Study of Natural Variations and Hybrid Detection in Populations of Alnus incana (L.) Moench and Alnus rohlenae Vít, Douda and Mandák

AB  - Landmark-based geometric morphometrics (GM) was used to examine, for the first time, spontaneous hybridization between Alnus incana (L.) Moench and Alnus rohlenae Vít, Douda and Mandák, and to assess inter- and intrapopulation variability in leaf shape, leaf size and venation in natural populations in Serbia (Western Balkans). Two geographically distant (30 km) and two close (1.2 km) populations were selected to examine hybridization. The variability in leaf shapes was assessed by canonical variate analysis and linear discriminant analysis performed on the symmetric component of variation. Covariation between the symmetric component of shape variation and the number of pairs of secondary leaf veins was investigated with partial least squares analysis. Static allometry was examined for the first time in the genus Alnus Mill. A higher proportion of A. incana leaves was classified as A. rohlenae in geographically close populations, which is in accordance with the hypothesis about spontaneous hybridization. No single leaf of A. rohlenae was classified as A. incana, indicating that putative hybrids can only be found in grey alder populations. This study demonstrates that GM is a powerful tool for species delimitation and hybrid detection in the genus Alnus and it can be used for preliminary screening in hybrid zones.

AB  - β-cyclodextrin (β-CD) is a good host for the encapsulation of fennel and basil essential oils
(FEO and BEO, respectively) and the formation of inclusion complexes (ICs) using the co-precipitation
method. According to the results of the GC/MS analysis conducted in this study, monoterpenes and
monoterpenoids were the dominant chemical groups in total FEO, while in BEO, these two groups
occurred along with sesquiterpenes and sesquiterpenoids. The presence of dominant compounds
from both EOs was validated using the FT-IR spectra of ICs, which indicated successful complexation.
Analyses conducted using SPME/GC-MS showed the continuous emission of volatiles over 24 h from
both ICs. Under SEM, particles of both ICs appeared to have a rectangular or rhomboid morphology
and few aggregates. The insecticidal properties of EOs and ICs with β-CD were tested on the
Colorado potato beetle (CPB) as a model pest. The inclusion complex of β-CD with FEO altered the
developmental dynamic and body mass of the CPB. The initial increase in the proteolytic activity of
CPB larvae fed with potato plants sprayed with ICs was not maintained for long, and the proteolytic
efficacy of treated larvae remained in line with that of the control larvae. Future investigations will
focus on manipulating the volume of EOs used and the treatment duration for optimal efficacy and
potential application.

AB  - In this study, the concentrations of metals and trace elements (As, Cd, Co, Cr, Cu, Hg, Ni,
Pb and Zn) were determined in the muscle tissue of adult roach and white bream at two different
sites in the Belgrade section of the Danube. Twenty-six fatty acids, consisting of nine saturated FA
(SFAs), seven monosaturated FA (MUFAs) and ten polysaturated FA (PUFAs), were identified. The
analysis of the concentration of metals and trace elements of the roach and white bream showed
species-specific differences in their bioaccumulation. Four of all elements analyzed (As, Hg, Ni and
Pb) correlated significantly with the changes in FA profiles in fish from both sampling sites, with the
exception of Cu, which correlated with the FA profile at the site before, and Zn, whose concentration
influenced the FA profile at the site after wastewater discharges. The lower PUFA content in the fish
from a site under higher environment pressure could indicate that the fish are stressed. The results
suggest that changes in lipid composition may be one of the protective mechanisms of cells to cope
with anthropogenic stressors.

Anomalies of Sponge Spicules: Exploring Links to Environmental Pollution

AB  - The aim of this study was to assess the frequency of spicule malformations in freshwater sponges in relation to selected environmental parameters of the streams and the presence of river pollutants. A total of 50 sponge samples were collected from ten rivers in Serbia. Selected parameters of the water varied considerably at every site where sponges were found. After spicule preparation, the samples were subjected to morphological analysis by light and scanning electron microscopy, and the number of anomalies were recorded (spicules with bulbous enlargements, sharply bent, bifurcated, scissor- and cross-like, and t-shaped). The frequencies and types of malformations within the analyzed specimens varied from 1 to 100 per 1000 spicules, with an average number of 12 per 1000. The main types of anomalies were single- and double-bent spicules. The highest number of anomalies was found in a specimen of Eunapius fragilis collected at Markovac (Velika Morava River), and the lowest number was found in a specimen of Ephydatia fluviatilis from Kanjiža (Tisa River). The sites with the lowest and the highest numbers of anomalies showed statistically significant differences in concentrations of ammonia, orthophosphates, sodium, chloride, manganese, and lead. This study indicates that several pollutants potentially affect the occurrence of spicule anomalies.

Survey 4 (JDS4) expedition. Three freshwater benthic species were used to estimate the quantity
of microplastics (MPs): Corbicula spp., Limnodrilus hoffmeisteri (Claparede, 1862), and Polypedilum
nubeculosum (Meigen, 1804). Following the kick and sweep technique, individuals were sampled using
a hand net or dredge. In order to estimate the number of MP particles/individual particles/g wet
body mass, the body mass and total length of all specimens were measured. Alkaline (Corbicula spp.
and L. hoffmaisteri) and enzymatic (P. nubeculosum) protocols were performed for tissue degradation.
All samples were filtered through glass microfiber filters (mesh size 0.5 μm). The particles were
photographed, measured, and counted. A total of 1904, 169, and 204 MPs were isolated from
Corbicula spp., L. hoffmaisteri, and P. nubeculosum, respectively. To confirm the chemical composition
of isolated MPs, a subsample of 46 particles of the fragmented particles from 14 sampling sites was
analysed via μ-ATR-FTIR spectroscopy analysis. The particles were characterised as polycarbonate
(PC), polyethylene terephthalate (PET), polypropylene–polyethylene copolymer (PP-PE), nylon
(polyamide-PA) and cellophane, with the domination of PET.
